The application of serum-free media in the production of monoclonal antibodies (MAbs) from hybridoma and recombinant myeloma cell lines for research and diagnostic purposes is rapidly growing. Typically, serum-containing media are used to culture these cell lines, however, growing concerns over serum supply and the potential for exogenous serum contamination has driven the transition to serum-free media.

Since each hybridoma or recombinant myeloma cell line is unique, it is necessary that the serum-free medium of choice offer nutrients for a variety of applications. In addition to this, unique requirements of some cell lines require specific media supplementation to be effective (e.g., cholesterol auxotrophic cell line, NS0).

HyClone CDM4NS0™ (SH30579, SH30578)
The Premier Chemically-Defined Medium for Therapeutic MAb Production in NS0 Cell
HyClone CDM4NS0 is a chemically-defined medium containing no animal derived components. This regulatory-friendly medium is developed through Metabolic Pathway Design™ to increase process yields in the manufacture of monoclonal antibodies (MAbs) using a variety of NS0 cell clones. HyClone CDM4NS0 requires no cholesterol or GS supplementation, as it has sufficient amounts to support NS0 cell culture already in the formulation. It has been successfully tested in a variety of culture systems, including T-flasks, shaker flasks and bioreactors using batch and fed-batch strategies.

HyClone CDM4NS0 contains Pluronic® F68, and does not contain L-glutamine and phenol red. HyClone CDM4NS0 is available in small and large-scale liquid volumes ranging from 500 mL PETE bottles to 500 L BioProcess Container™ Sytems (BPC®). It is also available in a cost-effective dry powdered formulation for on-site hydration in our unique HyNetics™ single-use production technology or in existing media formulation facilities.

HyClone CDM4MAb™ (SH30801, SH30802, SH30800)

HyClone CDM4MAb is a chemically-defined medium containing no animal derived components. This regulatory-friendly medium is developed through Metabolic Pathway Design™ to increase process yields for the manufacture of monoclonal antibodies for the therapeutic use in a variety of engineered hybridoma and recombinant myeloma cell lines.

HyClone CDM4MAb conatins Pluronic® F68 and does not contain phenol red. It is available with and without L-Glutamine.

HyClone SFM4MAb™ (SH30513, SH30391, SH30535)
The Premier Serum-Free Medium for Therapeutic MAb Manufacturing

This high performance cell culture medium is developed through Metabolic Pathway Design™ to increase the process yields for the industrial manufacture of human and humanized recombinant antibodies and antibody fragments for therapeutic use in a variety of engineered hybridoma and recombinant myeloma cell lines.

HyClone SFM4MAb is a low protein formulation and is optimized for downstream purification using Protein A, Protein G and other matrices to facilitate product recovery. It is formulated using HyClone’s proprietary lipid and phospholipid complexing process for enhanced stability and growth promotion of various cell types.

HyClone SFM4MAb contains synthetic cholesterol, L-Glutamine and Pluronic® F-68, and does not contain phenol red. It is also available without L-Glutamine to support the GS gene expression system.

HyClone SFM4MAb™-Utility (SH30382, SH30550)
The Preferred Serum-Free Medium for MAb Production in Research and Diagnostics

This versatile serum-free cell culture medium is developed through Metabolic Pathway Design™ to support the growth of multiple hybridoma cell types and production of a variety of immunoglobulins. It enables superior growth of many hybridomas and recombinant myelomas with minimal adaptation.

HyClone SFM4MAb-Utility is ideally suited for the cost-effective manufacture of monoclonal antibodies for academic and industrial research, genomics and proteomics, in vitro diagnostics, drug target screening and validation, and manufacturing of preclinical lots. It is formulated using HyClone’s proprietary lipid and phospholipid complexing process for enhanced stability and broad-spectrum growth promotion.

HyClone SFM4MAb-Utility contains cholesterol,and does not contain phenol red or Pluronic® F-68. This serum-free medium has been successfully tested in a variety of culture systems including T-fl asks, spinners, roller bottles and various bioreactors, including hollow fiber.

HyClone ADCF-MAb™ (SH30349, SH30547)
The Leading Animal Derived Component Free (ADCF) Medium for MAb Production

HyClone ADCF-MAb is a protein-free medium containing no animal derived components. This regulatory-friendly medium is developed through Metabolic Pathway Design™ to increase the process yields for the manufacture of antibodies and antibody fragments for therapeutic use in a variety of engineered hybridoma and recombinant myeloma cell lines.

HyClone ADCF-MAb contains L-Glutamine and Pluronic® F68, and does not contain phenol red. It is also available without L-Glutamine to support the GS gene expression system.

HyClone LS250™ (SH30555)
The Premier Animal Derived Component Free (ADCF) Lipid Supplement for NS0 Cells
HyClone LS250 is a chemically-defined, ADCF lipid supplement specifically developed through Metabolic Pathway Design™ to stimulate cell growth and monoclonal antibody (MAb) production in cholesterol auxotrophic NS0 cell lines. This 250X concentrated lipid supplement has been formulated using a proprietary lipid complexing process for enhanced stability and fi lterability. HyClone LS250 has been designed to supplement a variety of HyClone serum-free media, and may be used in the preparation of liquid media from dry powdered media.

HyClone LS1000™ (SH30554)
The Preferred Animal Derived Component Free (ADCF) Cholesterol Supplement
HyClone LS1000 is a chemically-defined, ADCF cholesterol supplement specifically designed to be added as a fed-batch supplement or to sterile liquid media at the time of use. This 1000X concentrated supplement has been formulated using a proprietary complexing process for enhanced cholesterol delivery.

In addition to these recently developed serum-free media, HyClone continues to support its early generation products, HyClone CCM1™, HyClone PF-MAb™ and HyClone SFX-MAb™. HyClone CCM1 is a serum-free medium for a variety of hybridoma and myeloma cell lines, and has been shown to work in a variety of alternative cell lines, including lymphocytes and epithelial cells. HyClone SFX-MAb is a serum-free medium for hybridoma and myeloma cell lines. HyClone PF-MAb is a 100X concentrate designed for use with RPMI-1640 and other basal formulations to reduce or eliminate serum dependency.
